Teaching at the Harvard Medical School for over 16 years, Dr. David Sinclair is a world-leading expert when it comes to aging, why we age, and the genetics that underpins the aging process.

His research has paved the way for cutting-edge bio-hacks on epigenetic reverse aging, and how certain enzymes react to different levels of NAD+, giving the public ways to fight against illnesses and cancer, among other aging ailments.

David Sinclair is also the author of “Lifespan: Why We Age – And Why We Don’t Have To,” a best-seller that talks about longevity, how we can expand our lifespans, and the proven research behind anti-aging methods.

What Supplements Does David Sinclair Take?

David Sinclair has made a point to refrain from recommending any supplement, instead suggesting to his readers to look for supplements from reputable companies with a high pure molecule percentage.

However, David has shared from his books and other public sources what he takes, and we’ve made a list that compiles all of this information. Unfortunately, as this isn’t directly from David, we cannot be certain whether his regimen has changed or if he takes additional supplements beyond this list.

As an example, David has mentioned in “Lifespan” that NMN and resveratrol are on his list of daily morning supplements, which he takes with yogurt.

We’ve compiled a list based on how often David mentions the supplements he takes, thus implying how important each supplement is to him and his regimen. This list includes information David has provided from interviews, books, and other public sources.

David Sinclair Supplements List

Dr. David Sinclair’s daily supplement stack, taken from various sources and have ranked them based on our view of their importance and efficacy in the Longevity space:

    1. Metformin: Taken in the morning, at around 800 mg
    2. NMN: Taken in the morning, at 1g
    3. Resveratrol: Taken in the morning as well, at 1g 
    4. Quercetin and Fisetin: Taken in the morning, at roughly 500 mg for each
    5. Spermidine: Taken in the morning at roughly 1 mg
    6. Trimethylglycine: Taken daily at roughly 500 to 1000 mg
    7. Vitamin D3: Taken daily, at around 4000 to 5000 IU
    8. Vitamin K2: Taken daily at around 180 to 360 mg
    9. Alpha-lipoic Acid: Amount taken currently unknown
    10. Coenzyme Q10: Amount taken currently unknown
    11. Low-dose Aspirin: Taken daily at roughly 83 mg
  • Statins: Taken daily at roughly 80 mg

  • Metformin

    According to David, the proven benefits of metformin are surprising and numerous. Going on to mention several studies in his book, “Lifespan,” David noted that Metformin was “particularly promising,” giving hope to longevity enthusiasts worldwide.

    Metformin, when given to diabetic patients, garnered results that could be applied to combating diseases, while also affecting the aging process. David also mentioned research where patients taking metformin had lower mortality rates compared to both diabetic and non-diabetic patients, along with an overall less amount of cardiovascular disease and cancer. No other anti-diabetic drug could produce similar results.

    Metformin, however, does have some side effects for some people, including diarrhea and abdominal discomfort when first introduced into the diet. After a few months, metformin has been known in some cases to reduce the body’s ability to absorb vitamin B12.

    Metformin is so effective because it causes our cells to repair and protect themselves due to the slight amount of damage metformin causes to the cells, specifically as a mitochondrial inhibitor.

    Due to this, David Sinclair never takes metformin on the days he exercises, as exercise creates a similar effect on the body’s cells, which could potentially put too much stress on the body if both are done at the same time.

    Metformin is also not recommended for those who are beyond a certain age threshold, such as being over the age of 65. This is because metformin could potentially cause too much stress on already aged cells.

    Dr. David Sinclair also mentions that the timing of metformin is incredibly important, as metformin can enhance the fasting state of the body. Our bodies automatically go into a fasting state while we sleep, so David takes metformin typically before bedtime at around 800 mg, once a day.

    Whilst Metformin is a very promising life-extension option, it is currently a medicine and therefore only available on prescription. Fortunately, there are natural molecules that have been found to have similar effects on the body and are not classified as a medicine.     Research suggests that NMN can affect many aspects of age-related diseases. One study found that mice that were given NMN had their NAD+ levels increase, leading to a decline in physiological diseases.

    David himself found the data promising, quoting in “Lifespan” that if a test subject was given anything with NMN in it, their NAD+ levels would rise by at least 25%, the equivalent of having fasted or exercised a considerable amount.

    NMN is imperative to the human body; It’s a molecule that enables proteins to properly function with a variety of jobs, such as repairing DNA and the epigenome.

    With 1000 mg being on the higher dosage amount, 250 or 500 mg is also a fair dosage amount to reap the benefits of NMN. This particular study showed promising results with just 250 mg daily.

    On page 150 of David’s book, he comments that many people ask him which is better, NMN or NR?  He goes on to say that his research suggests that NMN is the preferable choice out of the two, as certain health benefits aren’t seen in NR as they are in NMN when used in studies. However, NR has still proven to expand life, and while NMN is still undergoing testing, David isn’t able to give a definitive answer yet.

    David mentions in “Lifespan” that resveratrol, a stilbenoid acquired from grape skins, seems like a suitable candidate for cancer and cardiovascular disease prevention. Resveratrol is proven to have the ability to affect tumors, which can be related to other diseases with similar expressions in the body.

    Not only is resveratrol effective on its own, but David also believes that resveratrol’s full potential is unlocked with the help of NMN. This potential is found in two parts: activating the sirtuin genes (the protectors of the body’s DNA and epigenome) with resveratrol, while giving the genes energy with NMN.

    Dr Sinclair was central to the team that discovered the benefits of Resveratrol. In the clip he talks about the positive impacts it has on health and longevity.

    Quercetin and Fisetin

    Often referred to as “senolytics,” quercetin and fisetin are similar to each other as both share the ability to remove senescent cells from the body.

    Senescent cells, in particular, are cells that typically accumulate in older bodies, releasing byproducts that end up harming healthy cells.

    However, what makes these molecules stand out among others for life extension is the ability, as well as being a “senolytic”, to also reduce inflammation in the body.

    However, we believe the main reason why molecules like fisetin can extend lifespan is because of other effects than being a “senolytic”, such as reducing inflammation.

    David mentions in an interview that the studies around quercetin and fisetin are incredibly interesting for longevity, saying that mice who were given fisetin had their lifespans increase by at least 10%.

    David mentioned in a tweet that Spermidine, found in wheat germ, has proven research that encourages a healthy heart and extends life when used in studies with mice, given if the molecule is administered halfway through the mice’s life.

    Regarding people using spermidine for anti-aging, David goes on to mention that spermidine regularly consumed can be related to a lower risk of developing diseases, and a life extension of 5-7 years.

    Spermidine is found in a variety of foods, such as cheese, mushrooms, and soy, and is linked to age-related processes, such as autophagy.

    The studies around spermidine are also promising, showing that spermidine can affect the human body’s longevity and overall health.

    Spermidine is one of the newer additions to Dr Sinclair’s protocol. He talks about it in this short clip.

    Our spermidine contains 10mg of the active ingredient per gram - the highest potency on the market.

    TMG (Trimethylglycine or Betaine)

    In a podcast, David mentioned that taking TMG is recommended to avoid the possibility of NMN reducing methylation.

    Methyl groups are essential for DNA and proteins to function correctly. A high amount of NMN being processed in the body to increase NAD+ levels leads to a byproduct (nicotinamide) that has to be removed, which requires methyl groups.

    So to avoid a deficiency of methyl groups, TMG could be used to supplement the number of methyl groups in the body.

    Vitamin D3

    According to the research, Vitamin D3 (naturally found in fish and fish liver oils) extends life in proteins. It can also reduce the chances of developing an age-related disease, as a deficiency in vitamin D has been seen in case of the increased likelihood of type 2 diabetes, heart disease, and Alzheimer’s.

    Vitamin D is essential for activating genes that give your body the health and homeostasis it needs. Most governments, however, advise too low of an amount to take daily (under 1000 IU.) Many nutritionists recommend taking over 4000 IU per day and getting your vitamin D levels checked.

    Vitamin D is one of the most common vitamins taken and can be found everywhere very cost effectively. 

    Vitamin K2

    Vitamin K2 is a super nutrient for the body, aiding in bone, mitochondrial and vascular health. One study found that subjects with a higher daily dose of vitamin K2 were as much as 52% less likely to have calcified arteries, and 57% less likely to develop heart disease.

    Ideally, if you’re taking vitamin D, it is best to also take vitamin K2 with it as both work synergistically in the body together. The best dosage amount for vitamin K2 is around 180 to 360 mg daily.

    Vitamin K is less common than vitamin D but is still widely available

    Alpha-lipoic Acid (ALA)

    In recent interviews, Dr. David Sinclair did not specifically mention taking this supplement, so he may have discontinued use.

    Alpha-lipoic acid is known to be a powerful antioxidant, and there has been much discussion on using ALA to reduce aging.

    However, many studies have found the data to be inconclusive on using ALA for this means, as the results found little evidence in slowing down aging or extending life.

    Since ALA is an antioxidant, and new research suggests that antioxidants have the potential to cause an acceleration in aging, it’s not surprising to find studies where mice taking ALA have shorter lifespans.

    Coenzyme Q10 (coQ10)

    In recent interviews, Dr. David Sinclair did not specifically mention taking coQ10, so he may have discontinued use.

    CoQ10, similar to ALA, is an antioxidant that helps with the healthy functioning of mitochondria. However, the data is inconclusive about coQ10’s ability to lengthen lifespan.

    One study, in particular, demonstrated that in a control group with mice undergoing various lifespan studies, the mice’s mortality rate showed no significant changes with or without the administration of coQ10.

    Low-dose Aspirin

    David mentions in an interview that not only is low-dosage aspirin useful for cardiac-related disease prevention but is also useful for reducing certain types of cancer, such as colon cancer.

    However, the data is inconclusive as a recent study showed that, comparing two control groups: one using low-dose aspirin, and one using a placebo daily, found little difference between the two groups in terms of disease prevention. On the contrary, the results stated that older adults taking low-dose aspirin were more likely to experience a hemorrhage, and the likelihood of developing cardiovascular disease was not lowered.


    Due to some family history of cardiovascular illnesses, David mentioned in a podcast that he takes 80 mg of statins daily.

    Statins have the potential to lower the possibility of developing heart disease, however, there is some difference in opinion on whether statins are truly effective.

    If there is more of a likelihood of you getting a heart attack, some scientists suggest prescribing statins as preventative medicine, while other scientists suggest that statins should only be used after you’ve experienced a heart attack.

    Statins seem to heavily depend on a person’s genes and genetic make-up, as research suggests that some people react well to statins while others don’t seem to react at all.

    Statins are also not made equally as some can extend the lifespan of mice (such as simvastatin), while others are unable to reproduce the same effect. The side effects of statins are also fairly strong, including nerve pain and sore muscles, which can vary in strength depending on the type of statin.

    The side effects are understandable, as statins reduce the body’s levels of cholesterol, an important component of the body’s neuron and muscle cells. A reduction in cholesterol means that the body has less of a vital ingredient it needs to function properly.

    What Other Longevity Techniques Does David Sinclair Use?

    Supplements are only one side of a healthy life, and one should also include other aspects to reap the full benefits of longevity practices. David shares in “Lifespan” how he fully lives the longevity lifestyle through three methods: proper nutrition, exercise, and health monitoring.


    Quoting “Lifespan” on page 303, David avoids sugars and desserts as much as possible, and typically skips one meal a day. He also avoids microwaved plastic and doesn’t smoke. And regarding his ratio of vegetables to meat, he aims to eat more vegetables and as little meat as possible.


    David tries to include as many steps in his day as possible and takes the stairs when he has the chance. And on the weekends, he goes with his son to the gym to lift weights.

    Health tracking

    After every few months, David has a professional come to take a blood sample, analyzing it for multiple biomarkers. Anything that is outside of the optimal range, David then adjusts with food or exercise.


    David Sinclair’s stack of supplements is effective for increasing one’s lifespan, but the doctor has stated that his personal stack may not work for others, as everyone’s genes are unique. 

    Different supplements also have different effects, such as NMN with proven life extension traits in mice plus a variety of human trials underway, making it a longevity supplement, while others improve the body’s basic functions, making them health supplements. Taking this into consideration, it’s possible to consider David’s expertise while making your own anti-aging supplement stack for a longer, healthier life.
